ANCHORAGE, Alaska (AP) ? An Alaska State Trooper helicopter carrying two troopers and a rescued snowmobiler crashed Saturday night in the south-central part of the state, and no survivors have been found, an agency spokeswoman said.
Trooper spokeswoman Megan Peters said the crash site was spotted Sunday, but she could not immediately confirm that the three on board were killed.
Wreckage of the helicopter burned, but Peters said it was not known how the fire started or how long it lasted.
The trooper helicopter was on a mission to pick up a snowmobiler stranded near Larson Lake 7 miles east of Talkeetna in the Matanuska-Susitna Borough.
The helicopter picked up a trooper and began a search, spotting the snowmobile and landing just before 10 p.m. It took off with the snowmobiler and was supposed to meet medics near Talkeetna but did not arrive.
The National Weather Service said the temperature in Talkeetna Sunday night was 34 with light rain that turned into snow at about 11 p.m., but that visibility was 10 miles.
Peters said it's not known if weather was a factor in the crash. The National Transportation Safety Board will investigate.
Alaska Air National Guard helicopter spokeswoman Kalei Rupp said fog in Anchorage and mandatory crew rest kept an agency helicopter from launching until 5 a.m.
A guard helicopter spotted the wreckage at 9:20 a.m. Sunday and landed.
Talkeetna is a community of nearly 900 about 80 miles north of Anchorage. It's the primary departure point for air taxis taking climbers to Mount McKinley.
The crash site was near the south end of Larson Lake.
The Alaska State Troopers have had at least one officer die in a helicopter crash during a search and rescue mission.
Trooper John David Stimson was on a chartered helicopter in 1983 that was attempting to reach a downed pilot in a blizzard. The chartered helicopter crashed near Cordova.
Stimson survived and assisted pilot Gary Wiltrout, who had suffered a broken back. However, when rescuers reached the crash site after 24 hours, the 11-year trooper had died of exposure. Wiltrout survived.

Most commonly associated with such maladies as "mad cow disease" and Creutzfeldt-Jakob disease, prions are increasingly recognized for their ability to induce potentially beneficial traits in a variety of organisms, yeast chief among them.
Now a team of scientists has added markedly to the job description of prions as agents of change, identifying a prion capable of triggering a transition in yeast from its conventional single-celled form to a cooperative, multicellular structure. This change, which appears to improve yeast's chances for survival in the face of hostile environmental conditions, is an epigenetic phenomenon?a heritable alteration brought about without any change to the organism's underlying genome.
This latest finding, reported in the March 28 issue of the journalCell, has its origins in work begun several years ago in the lab of Whitehead Institute Member Susan Lindquist. In 2009, Randal Halfmann, then a graduate student in Lindquist's lab, identified dozens of proteins in yeast that have the ability to form prions. That research greatly expanded the known universe of prion elements in yeast, but it failed to answer a key question: What function, if any, do these prions actually have?
In search of an answer, Halfmann, now a fellow the University of Texas Southwestern Medical Center, and colleagues in the Lindquist lab attempted to exploit the fact that several of the prion-forming proteins they had identified acted to modify transcription of yeast genes. It stood to reason that if they could identify which genes were being regulated, they might be able to determine the prions' function.
"We looked at the five transcriptional regulators that are known to be prions in yeast, and we found that in fact, only one gene in the entire yeast genome was regulated by all five transcription factors," says Halfmann.
That gene, as it turns out, was FLO11, a key player in multicellularity in yeast. Indeed changes in FLO11 expression have been shown to act as a toggle, switching yeast from spherical to filamentous form. Halfmann notes that FLO11, which has been shown to be regulated by epigenetic elements, is also highly responsive to environmental stress. Knowing that the prion form of a protein is essentially a misfolded form of that protein, and that stressful conditions increase the frequency of protein misfolding and prion formation, the scientists began to consider the possibility that the prions themselves might be among the epigenetic switches influencing the activity of FLO11.
The group focused on one transcription factor known as mot3, finding that yeast cells containing the prion form of this factor, [MOT3+], acquired a variety of multicellular growth forms known to require FLO11 expression. This was a clear indication that prion formation was causing the differentiation of the cells and their subsequent cooperation. But what about the stress aspect of the hypothesis?
By testing yeast cells against a variety of stressors, the scientists discovered that exposure to a concentration of ethanol akin to that occurring naturally during fermentation increased [MOT3+] formation by a factor of 10.They also found that as the cells exposed to ethanol shifted their metabolism to burn surrounding oxygen through respiration, the prions reverted to their non-prion conformation, [mot3-], and the yeast returned to the unicellular state. In essence, prion formation drove a shift to multicellularity, helping the yeast to ride out the ethanol storm.
"What we have in the end is two sequential environmental changes that are turning on a heritable epigenetic element and then turning it off," says Halfmann. "And between those two changes, the prion is causing the cells to acquire a multicellular growth form that we think is actually important for their survival."
Lindquist, who is also a Howard Hughes Medical Institute investigator, has long argued that prions have played a vital role in yeast evolution and has amassed a body of strong supporting evidence.
"We see them as part of a bet-hedging strategy that allows the yeast to alter their biological properties quickly when their environments turn unfavorable," Lindquist says. She also theorizes that prions may play such roles beyond yeast, and her lab intends to take similar approaches in the hunt for prions and prion-like mechanisms that are potentially beneficial in other organisms.
For Lindquist lab postdoctoral scientist Alex Lancaster, who is also an author of the new Cellpaper, these latest findings hint at a potentially novel approach to understanding basic mechanisms underlying the complexities of human diseases, including cancer, whose hallmarks include protein misfolding, epigenetic alterations, metabolic aberrations, and myriad changes in cell state, type, and function. Lancaster likens the opportunity to that of opening a black box.
"It's exciting to think that this could become another tool in the toolbox in the study of multicellularity," Lancaster says. "We know that some tumors are a heterogeneous population of cells and we know that tumor cells can evolve within in their environments to help ensure their own survival. This system could help us further understand the role of epigenetic inheritance within tumors and how it might be influencing cell-cell interactions and even affecting the effectiveness of drug therapies."

WASHINGTON (AP) ? After sweating through a pair of edge-of-your-seat comebacks, Marquette's first Sweet 16 victory in a decade was as straight and smooth as the 15-foot step-back jumper that Vander Blue nailed at the end of the first half.
It helped that the Golden Eagles ran into an out-of-sorts Miami team that, in an echo of its bus ride to the Verizon Center, was able to make as much headway as a frustrated commuter in rush-hour traffic.
Marquette is in the Elite Eight for the first time since 2003, getting there with an emphatic 71-61 win over Miami on Thursday night. The Golden Eagles were never threatened after taking a double-digit lead in the first half, quite the contrast from their rallies that beat Davidson by one and Butler by two earlier in the NCAA tournament.
"It's fantastic. It feels good not to have to worry about, are you going to lose on a last-second shot or are you going to win on a last-second shot?" said Jamil Wilson, who had 16 points and eight rebounds. "To have a cushion like that, these guys played with tremendous heart, and we did it all game."
Blue, who made the shot that beat Davidson and led the comeback against Butler, finished with 14 points. He wasn't Marquette's leading scorer, but his offensive and defensive energy pushed the Golden Eagles to a big lead early. It's a good thing he got his buzzer-beater before halftime ? for a change, Marquette didn't need one at the end of the game.
"We're so used to people not giving us credit. ... That fuels our fire," Blue said.
The third-seeded Golden Eagles (26-8) will face Big East rival Syracuse, the No. 4 seed, in the East Regional final on Saturday, aiming for a spot in the Final Foul for the first time since the 2003 team led by current Miami Heat star Dwyane Wade. Marquette was knocked out in the round of 16 the past two years.

Syracuse beat top-seeded Indiana 61-50 in the other East Regional semifinal.
This one wasn't hard to decipher. Marquette could shoot; Miami couldn't. The Hurricanes (29-7) had sentiment on their side, returning to the arena where coach Jim Larranaga led mid-major George Mason to the Final Four seven years ago, but they made only 35 percent of their field goals and missed 18 of 26 3-pointers.
"You ever have days where you're just out of sync or things just don't run along smoothly?" Larranaga said. "Almost like our trip over here. Our hotel is a mile and a half, it took us 45 minutes to get here. We had to go on nine different streets, weaving our way in and out of traffic and everything. And that's the way it seemed on the court. We were trying to find our way and never could. Never could get in rhythm offensively, and defensively. I don't think we communicated like we have been doing all season long."
Shane Larkin scored 14 points to lead the No. 2 seed Hurricanes, whose NCAA run to the round of 16 matched the best in school history.
"I think what we did this year was lay a foundation of what the program could be like," Larranaga said. "We're not anywhere near where I would like to be."
Marquette, meanwhile, shot 54 percent, a stark turnaround from its 38 percent rate from the first two games in the tournament. Davante Gardner added 14 points, with 12 coming in the second half when the Golden Eagles were comfortably ahead.
Comfortable being a relative term. Coach Buzz Williams, who relishes the Golden Eagles' underdog status, hardly seemed to know how to take such an easy win. He didn't look or sound like a winning coach afterward.
"Because of my path to this point, I do have an edge, and I probably need to have better wisdom as to how to handle that edge," Williams said. "But it's really delicate because our edge is why we win."
Blue got going when he picked off a pass and converted the steal into a one-handed jam to give Marquette an 8-4 lead. His running one-hander made it 12-4. Blue and Junior Cadougan forced a steal, getting Larkin to commit his second foul in the process.
Blue ended the half with an exclamation point, hitting the jumper just before the horn to give Marquette a 29-16 lead at the break. He drained the shot, strutted backward downcourt, cocked his right arm and gave Wilson a chest bump.
Meanwhile, the Hurricanes couldn't sink anything. They started 2 for 12, including 0 for 6 from 3-point range, and Larkin's 3-pointer more than 11 minutes into the game was the first Hurricanes field goal scored by anyone other than Kenny Kadji.
In the second half, Blue's basket with 10:03 to play gave Marquette a 51-30 lead. The Hurricanes, who by then had started to press full court, then put together their best sequence of the night, a 7-0 run that cut the lead to 14 with 8? minutes left.
But Wilson's dunk and Gardner's inside basket stretched the lead back to 18. Gardner became the scene-stealer late, thumping his chest to the Marquette fans after a dunk in the final four minutes.
The Hurricanes played without backup center Reggie Johnson, who had surgery Tuesday for a minor knee injury. Johnson was averaging seven rebounds, but he would have helped only if he could've put the ball in the basket.
"There are only two things you have to do in basketball ? one, put the ball in the basket, two, stop the other team from putting the ball in the basket. We weren't able to do either," Larranaga said.

FRESNO, Calif. (AP) ? Former "Bachelorette" contestant Kasey Kahl will avoid jail time after pleading no contest to battery stemming from a bar fight in California.
The Fresno Bee reports (http://bit.ly/11V25k4 ) Kahl entered the plea Thursday to a felony count of battery causing bodily injury. An assault count was dismissed.
The 30-year-old Clovis man was sentenced to 180 days of community service.
Kahl was accused of breaking a man's nose during a fight last year in Fresno. His attorney, Gerald Schwab, says his client acted in self-defense.
Kahl was a contestant on ABC reality TV show's "The Bachelorette" in 2010.

I have a very large app built on my desktop. I want to continue working on it on my laptop. I tried out Team Server and uploaded the solution. I then download the solution to my laptop.
I cannot build it on my laptop. I get 75 errors for Resources is not a member of my
I also tried just directly copying the full folder from my desktop but its the same.
I have googled this for weeks and tried every suggestion on the net i can find.
Does anyone know where i start with this? What info can I give to help?

Is the directory correct in both cases? In particular, are the paths identical between your main machine and the laptop? If not, are they logically equivalent? ie. at least the relative paths should be the same, or point to actual instances of the .Designer.vb files.
Within the .Designer.vb file, there should be a line similar (but obviously not the same as):
BEIRUT (AP) ? Mortar shells crashed into an outdoor cafe at Damascus University on Thursday, killing at least 10 students in the deadliest of a rising number of mortar attacks in the heart of the Syrian capital.
The strikes have escalated as rebels fighting to topple President Bashar Assad try to enter the city, terrifying civilians whose support the opposition needs to advance its cause.
It was unclear who fired the rounds. The government blamed "terrorists," its blanket term for those fighting Assad's regime. Anti-regime activists accused the regime of staging the attack to turn civilians against the rebels.
Mortar strikes on Damascus are relatively new in Syria's crisis, which began in March 2011 with protests calling for Assad's ouster, then evolved into a civil war. The U.N. says more than 70,000 have been killed in the conflict.
Since last month, mortar shells have hit previously safe parts of the capital with increasing frequency. The near-daily strikes have frightened residents, and many have begun to avoid open areas and put plastic on their windows to help block flying glass from an explosion or shrapnel.
Some shells appear aimed at government targets, such as one of Assad's palaces and the general command of the Syrian army. Others have hit near civilian targets, including the Sheraton Hotel and a soccer stadium, both on the city's west side. Mortar shells also have struck in areas to the east, like the Christian neighborhood of Bab Touma.
Thursday's strike was the deadliest yet.
State-run Al-Ikhbariya TV showed video of the university cafe where blood pooled on tiles and plastic chairs and pens and eyeglasses littered the area. Later video showed people being treated in a hospital, including a woman with white bandages around her head and a man whose back was peppered with shrapnel wounds.
The dining facility belongs to the Faculty of Architecture in Damascus's central Baramkeh district.
State TV said 15 people were killed in the strike, but the official news agency, SANA, put the death toll at 10 and said dozens were wounded. It also reported three other mortar strikes nearby.
The opposition activist group, the Britain-based Syrian Observatory for Human Rights, put the death toll at 13.
Similar mortar attacks on Tuesday killed at least three people and wounded dozens. Six people were killed by mortar shells in different parts of the city on March 11.
"No one anywhere in the world can imagine a more criminal act than this," SANA quoted Amer al-Mardini, the president of the university, as saying. He said he hoped the wounded would heal quickly and "resume their studies as soon as possible."
Anti-regime activists accused the regime of launching the attack to tarnish the opposition's image.
Elizabeth O'Bagy, who studies the Syrian rebels at the Institute for the Study of War, said it was not possible to determine who was behind the attack, but it appeared to fit the regime's pattern of escalation. In other aspects of the war, such as the use of airstrikes or Scud missiles, the regime has gone from trying to target rebels to more indiscriminate attacks on civilians, she said.
"Because of the fact that it does follow regime behavior, it is more likely to be a regime attack," she said, while acknowledging it could also have been a rebel misfire.
Rebels have established footholds in a number of Damascus suburbs but have only been able to push into limited areas in the south and northeast parts of the capital. The government has retained its grip of downtown Damascus, although the mortar strikes have deepened fear among many residents that they will soon see the violence that has damaged many other Syrian cities.
Thursday was not the first time Syria's universities have been targeted. On Jan. 15, twin blasts hit Aleppo University, killing more than 80 people. The opposition said the regime had bombed the university, while the government accused rebels of striking it with rockets.
Also Thursday, Ghassan Hitto, the newly elected prime minister of the main opposition bloc, said he was reviewing candidates for a planned rebel interim government. It will be a service-oriented administration with nine to 12 ministries and will be based inside Syria, Hitto said during a meeting with Syrian expats in Qatar.
In Washington, State Department spokesman Victoria Nuland said the head of the opposition Syrian National Coalition, Mouaz al-Khatib, has decided to continue serving until his six-month term ends in May. Al-Khatib resigned on March 24, citing frustration with the group and its level of international support. The Coalition rejected his resignation.
Syria's conflict threatens to destabilize neighboring countries, where more than 1 million refugees have fled to escape the violence.
In Jordan, on Syria's southern border, a riot broke out Thursday in a refugee camp after Jordanian authorities refused to let buses full of refugees return to Syria because of violence over the border. U.N. refugee liaison Ali Bibi said it was unclear how many refugees were involved in the melee at the Zaatari camp, but no one was injured.
To the north, Turkey denied reports that it was deporting hundreds of Syrian refugees for rioting on Wednesday in a camp in Akcakale after a fire killed a 7-year-old child. A camp official said local authorities identified 300 people involved in the uprising and prepared to deport them, but the government stopped them.
A Foreign Ministry official said 100 refugees asked to leave the camp and return to Syria on their own.
The U.N. refugee agency did not confirm the reports, but said it was concerned about possible deportations of refugees.
In Israel, on Syria's southeastern border, the military said it was beefing up medical teams along the border because of several cases of wounded Syrians crossing the frontier for medical care. Eleven Syrians have been treated in Israeli hospitals, including one who died from his wounds on Wednesday, a military official said. Others returned home after their conditions improved.
All officials spoke on condition of anonymity because they were not authorized to brief reporters.

WASHINGTON (AP) ? Former Secretary of State Hillary Clinton is returning to the public stage with her first speaking events since leaving the Obama administration.
Clinton is scheduled to speak Tuesday at the Vital Voices Global Leadership Awards in Washington, D.C., an event honoring women leaders around the world. A Clinton spokesman says it will mark her first public speaking event since leaving the State Department.
On April 5, the former New York senator and first lady will address the Women in the World Summit in New York, an annual event focusing on challenges faced by women and girls around the world. Clinton has addressed both organizations previously.
Clinton is scheduled to address The Economic Club of Grand Rapids, Mich., on June 17, organizers said. The event will be closed to the press.
Mar. 28, 2013 ? A widely prescribed type 2 diabetes drug slows down the aging process by mimicking the effects of dieting, according to a study published today using worms to investigate how the drug works.
Following a calorie-restricted diet has been shown to improve health in later life and extend lifespan in a number of animals, ranging from the simple worm to rhesus monkeys. The type 2 diabetes drug metformin has been found to have similar effects in animals but until now it was not clear exactly how the drug delays the aging process.
Researchers supported by the Wellcome Trust and Medical Research Council looked at the effects of metformin on C. elegans worms that were grown in the presence of E. coli bacteria, a relationship similar to that which humans have with the 'healthy' bacteria in our gut. They found that the worms treated with metformin lived longer only when the E. coli strain they were cultured with was sensitive to the drug.
Dr Filipe Cabreiro from the Institute of Healthy aging at UCL, who led the research, explains: "Overall, treatment with metformin adds up to 6 days of life for the worm which is equivalent to around a third of its normal lifespan. It seems to work by altering metabolism in the bacteria that live in the worm, which in turn limits the nutrients that are available to the worm host and has a similar effect to restricting the diet."
Bacteria living in the gut have an important role in helping the host organism to digest and extract nutrition from food. Defects in gut bacteria have been linked to metabolic diseases such as obesity, diabetes, inflammatory bowel disease and cancer. It has also been suggested that gut bacteria may have an impact on the aging process, but this is the first study to suggest a mechanism for how this works.
The team used strains of E. coli with defects in genes that are linked to metabolism and tweaked the levels of nutrients available to tease out which metabolic pathways might be affected by the drug. They found that treatment with metformin disrupted the bacteria's ability to metabolise folate, a type of B-vitamin, and methionine, one of the building blocks of proteins. This limits the nutrients that are available to the worm and mimics the effects of dietary restriction to enable the worms to live longer.
However, when they added an excess of sugar to the diet, the team found that the life-extending effects of metformin were cancelled out. As the drug is used as a treatment for diabetes caused by elevated glucose levels in the blood, this finding is particular relevant for understanding how the drug works in people.
Professor David Gems, who directed the study, said: "We don't know from this study whether metformin has any effect on human aging. The more interesting finding is the suggestion that drugs that alter bacteria in the gut could give us a new way of treating or preventing metabolic diseases like obesity and diabetes."
Metformin is currently one of the most widely prescribed drugs and the findings should help to inform how it is used in patients.

OTTAWA (Reuters) - Canada's economy bounced back in January with 0.2 percent growth following the weakest two quarters since the 2008-09 recession thanks to solid manufacturing, mining and an end to a work stoppage in professional ice hockey, Statistics Canada said on Thursday.
Analysts surveyed by Reuters had expected a 0.1 percent increase in monthly gross domestic product following a 0.2 percent decline in December.
The data suggests the economy is starting the year on a more solid footing after a disappointing 0.6 percent annualized growth in the fourth quarter.
The Bank of Canada has projected a hefty 2.3 percent growth in the first three months of 2013, although it may revise that outlook in its quarterly update on April 17.
The Canadian dollar hit C$1.0145 versus the U.S. dollar, stronger than C$1.0157, or 98.45 U.S. cents immediately before the data. But the currency quickly gave back all of those gains.
In a separate report, Statscan said Canadian industrial product prices grew 1.4 percent in February from January, the biggest jump since June 2008 as prices for petroleum, coal and other commodities charged higher.
Manufacturing contributed the most to the rebound, expanding 1.2 percent on gains in both durable and non-durable goods. Producers of fabricated metals and wood products showed gains while the transportation equipment industry softened.
Statscan also noted strength in mining, quarrying and oil and gas extraction, which increased 0.2 percent, and in wholesale trade, up 0.7 percent. The arts and entertainment sector got a one-time boost of 4.1 percent after owners and players in the National Hockey League reached a deal to end their labor dispute and begin a delayed hockey season.
Industries that shrank in January included agriculture and forestry, construction, and finance and insurance.

(Reuters) - A federal magistrate judge has recommended the dismissal of a civil lawsuit in which Paul Ceglia, a former wood pellet salesman, claimed a huge ownership stake in Facebook Inc.
U.S. Magistrate Judge Leslie Foschio in Buffalo, New York, said on Tuesday that there is clear and convincing evidence that a 2003 contract under which Ceglia claimed a one-half interest in the social media company is a "recently created fabrication."
In October, Ceglia was criminally charged by federal prosecutors with mail and wire fraud in connection with the alleged forgery of documents related to Facebook.
Paul Argentieri, a lawyer for Ceglia, was not immediately available on Tuesday for comment.
In his civil lawsuit against Facebook and its chief executive, Mark Zuckerberg, Ceglia had claimed that a 2003 contract he signed with Zuckerberg, then a Harvard University freshman, entitled him to the stake.
Though Zuckerberg had done programming work for Ceglia's company StreetFax.com, Facebook maintained that Zuckerberg's agreement involving that entity, which did not entitle Ceglia to a big Facebook stake, was the real contract between the two.
In a 155-page recommendation, Foschio said Ceglia's arguments largely consisted of "self-defeating inconsistencies" that established the "fraudulent nature" of his claims.
"Plaintiff has utterly failed to rebut the plethora of evidence establishing that it is highly probable and reasonable the StreetFax document was the operative contract that governed the business relationship between plaintiff and Zuckerberg," the judge wrote.
Foschio also said it is "highly probable and reasonably certain" that the contract Ceglia said was real was "fabricated for the express purpose of filing the instant action."
The case now goes to U.S. District Judge Richard Arcara, who will decide whether to approve Foschio's recommendation.
Colin Stretch, deputy general counsel of Facebook, said in a statement that Foschio's recommendation "confirms what we have said from day one: this lawsuit is an inexcusable fraud based on forged documents."
The civil case is Ceglia v. Zuckerberg et al, U.S. District Court, Western District of New York, No. 10-00569.

Too much stress can have serious effects on your body and health, but alleviating it isn't as simple as just not doing the things that stress you out. Over at The Harvard Business Review, Heidi Grant Halvorson, Ph.D. (who's work you've seen here before) explains that sometimes it's your approach to stressful events and chronic stressors that matters more than the way you handle them after the fact. We focus frequently on coping mechanisms that help you minimize the effects of stress, but she points out that with a healthy mindset and approach to stressful situations?one that looks at the stress you experience as something that can strengthen you instead of harm you?you'll weather the stressful storm a bit better.
The full study, published in the Journal of Personality and Social Psychology, turned up some surprising results:
in their studies, Crum and colleagues began by identifying stress mindsets among a group of nearly 400 employees of an international financial institution. They found that those employees who had stress-is-enhancing mindsets (compared to stress-is-debilitating) reported having better health, greater life satisfaction, and superior work performance.
That's already rather amazing, but here's the best part - your mindset can also change! If you have been living with a stress-is-debilitating mindset (like most of us), you don't have to be stuck with it. A subset of the 400 employees in the aforementioned study were shown a series of three-minute videos over the course of the following week, illustrating either the enhancing or debilitating effects of stress on health, performance, and personal growth. Those in the stress-is-enhancing group (i.e., the lucky ones) reported significant increases in both well-being and work performance.
Now to be clear, the study isn't saying that stress doesn't stress you out, or that too much stress is somehow healthy. What the researchers point out is that your attitude and approach matter a lot, almost as much as knowing how to cope with the stress afterward. In fact, take the study results with a healthy dose of skepticism. The results are preliminary, and just one study in the face of a mountain of research about stress. However, there's no harm (and everything good) about changing your mindset towards the things that stress you out, and if there are tangible health and lifestyle benefits to doing so (and they're backed up by some research), then by all means, do it.
